DTT for Protein and Reagent Production
DTT is used where a protein-processing formulation requires a reducing environment. Its applications include isolation of proteins and enzymes, reduction before selected protein-modification reactions, and manufacture of aqueous reducing reagents. A reported redox potential is approximately −0.33 V at pH 7.
The reduction of structural disulfide bonds can unfold a protein. In other systems, protection of oxidation-sensitive thiols helps preserve the required enzyme activity.
Dithiothreitol Technical Profile
Commercial reference profile; supply specifications remain specific to the offered grade.
| Test item | Commercial reference data |
|---|---|
| Assay, iodometric titration | ≥99.0% |
| Corrected melting point | 42 ± 2°C |
| Loss on drying | <0.50% |
| Oxidized DTT | <0.50% |
| Appearance of a 10% aqueous solution | Clear and colourless |
| Infrared identification | Consistent with the assigned structure |
| Thin-layer chromatography | Single spot |
The same reference grade includes UV absorbance limits of less than 0.067 at 280 nm and less than 0.050 at 305 nm for a 1% solution. At a concentration of 0.02 M, its absorbance limit at 283 nm is less than 0.040.
DTT is sensitive to oxidation during storage and solution preparation. The oxidized fraction is included in the commercial reference profile alongside the reducing-agent assay and solution-appearance limits.
Commercial Applications
Protein Isolation and Processing
DTT provides reducing conditions during selected protein isolation and processing operations. It protects free thiols from oxidation and reduces disulfide bonds during denaturing steps. Reduction can unfold proteins whose native structures depend on those bonds; this effect is relevant to processes that deliberately require denatured material.
Enzyme Isolation and Formulation
DTT is used in the isolation of enzymes with oxidation-sensitive sulfhydryl groups. In suitable systems, it protects these groups during processing and can restore activity affected by reversible thiol oxidation. Structural disulfide reduction can adversely affect enzymes that depend on those bonds for their active conformation.
Manufacture of Reducing Reagents
DTT acts as the reducing component in commercial protein-processing reagents and buffers. Its water solubility supports aqueous formulation, while the solid material allows weighed addition during batch manufacture. Solution clarity and reducing activity are relevant to the finished reagent, particularly in products with optical measurement requirements. DTT solutions are susceptible to oxidation during storage and air exposure. Their reducing activity is also affected by pH, temperature and contact time.
Reduction Before Protein Conjugation
DTT can generate accessible protein thiols before selected conjugation reactions. The reduction step precedes the subsequent thiol-reactive coupling stage. Residual DTT reacts with maleimide groups and competes with the intended protein thiols, so excess DTT is removed before maleimide coupling. Depending on the established process, this may involve desalting or buffer exchange.
Packaging and Storage
Bulk commercial packaging includes 25 kg bags or fibre drums. These are reference pack formats; the actual net weight and packaging configuration apply to the offered material and order.
DTT is sensitive to air, moisture and heat. One established commercial grade specifies storage at 2–8°C under nitrogen. Protection against moisture and oxygen remains relevant after the outer transport packaging is opened. The supplied grade’s label and safety documentation determine its storage and handling conditions.
With a reference melting point near 42°C, DTT is susceptible to changes in physical form under warm conditions. Temperature protection during shipment is arranged for the supplied grade and route.
